Rat S100 Calcium Binding Protein A8 ELISA Kit

This sandwich ELISA from Antibodies.com is intended for the in vitro quantitative determination of rat S100 Calcium Binding Protein A8 (also referred to as MRP8) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The microtiter plate is pre-coated with a capture antibody specific for the target. Standards and samples are added to the wells and any target present is bound by the immobilised antibody; following washing, a biotinylated anti-target antibody binds the captured analyte and an HRP-avidin conjugate is then introduced. TMB substrate is added to visualise the HRP enzymatic reaction, producing a blue product that becomes yellow after addition of the acidic stop solution. The intensity of the yellow colour, read at 450 nm, is proportional to the amount of target captured, and the concentration is calculated by reference to the standard curve.

The assay covers a product range of 0.937–60 ng/ml with a sensitivity of <0.39 ng/ml and is completed in approximately 4 h 30 min on a colorimetric microplate reader. Spike-and-recovery has been validated in serum (range 80%–102%, average 91%, n=5), EDTA plasma (81%–101%, average 91%, n=5) and heparin plasma (80%–89%, average 84%, n=5). Dilution linearity has been demonstrated at 1:2, 1:4, 1:8 and 1:16 across serum (74%–107%), EDTA plasma (79%–108%) and heparin plasma (82%–105%).
