From Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of rat Retinoic Acid Receptor alpha in tissue homogenates or other biological fluids, with reactivity confirmed for rat samples. The assay employs the sandwich enzyme immunoassay technique, using an antibody specific for Retinoic Acid Receptor alpha pre-coated onto a pre-coated microplate of 12 x 8 well strips. Standards and test samples are added into the wells, where the target is captured by the immobilised antibody; bound analyte is then detected using a Biotinylated Anti-Retinoic Acid Receptor alpha Antibody, followed by HRP-Avidin conjugate and TMB substrate. The enzymatic reaction produces a blue-coloured product that turns yellow upon addition of acidic stop solution, and absorbance is read at 450 nm on a colorimetric microplate reader.

Reported performance figures include a sensitivity of 0.114 ng/ml and an assay range of 0.312-20 ng/ml, with a total assay time of 4h 30m. In validation across sample matrices (n=5), average recovery was 91% in serum (range 80-102%), 90% in EDTA plasma (range 81-100%), and 84% in heparin plasma (range 80-89%). Linearity on serial dilution was reported as follows: serum 87-91% (1:2), 87-107% (1:4), 74-101% (1:8), 92-97% (1:16); EDTA plasma 90-105% (1:2), 84-101% (1:4), 90-101% (1:8), 79-108% (1:16); heparin plasma 84-95% (1:2), 92-105% (1:4), 82-105% (1:8), 89-91% (1:16). Kit components should be stored at +4°C or -20°C as instructed, and the product must not be used past its expiration date.
