Rat REST ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) designed for the in vitro quantitative determination of rat REST/NRSF in serum, plasma, tissue homogenates, and other biological fluids. The kit is supplied by Antibodies.com and is reactive with rat samples, with no reported cross-reactivity with other analogs or variants. The microtiter plate is pre-coated with a capture antibody specific for REST/NRSF, and standards or samples are added to the wells where the target binds. Following incubation and washing, a biotinylated detection antibody is added to form an antibody-antigen-antibody sandwich complex. An HRP-Streptavidin conjugate and TMB substrate are then introduced to produce a blue product that turns yellow upon addition of the acidic stop solution. The intensity of the yellow colour is read at 450 nm and is directly proportional to the concentration of REST/NRSF.

The standard curve covers a product range of 31.25-2000 pg/mL, with a sensitivity of 18.75 pg/mL. Serum and plasma samples should be diluted at least 1:2 with Sample Dilution Buffer prior to testing, with validated dilutions for healthy serum and plasma (1:2) and rat brain tissue lysate (1:2). Recovery averages are reported as 96% for serum, 95% for EDTA plasma, and 97% for heparin plasma. The total assay time is 4 hours, and absorbance is read using a colorimetric microplate reader.

The kit is shipped at +4°C, and components should be stored at +4°C or -20°C as instructed in the protocol manual. The kit should not be used past its expiration date, and freeze/thaw cycles should be avoided.
