Rat Receptor Interacting Serine Threonine Kinase 3 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of rat Receptor Interacting Serine Threonine Kinase 3 (RIP3) in tissue homogenates, cell lysates, or other biological fluids. The plate is pre-coated with an antibody specific for RIP3; standards and test samples are added and RIP3 present in each sample is captured by the immobilised antibody. Following a wash step, a Biotinylated Anti-RIP3 Antibody is added, followed by an HRP-Avidin conjugate. Signal is developed using TMB substrate, producing a blue product that turns yellow upon addition of acidic stop solution; absorbance is read at 450 nm in a microplate reader and the RIP3 concentration is calculated against the standard curve.

The assay has a stated range of 0.156–10 ng/ml, a sensitivity of <0.055 ng/ml, and a total assay time of 4 h 30 min. Validation data supplied with the kit report an average recovery of 91% (range 80–102%) in serum (n=5), 90% (range 81–100%) in EDTA plasma (n=5), and 84% (range 80–89%) in heparin plasma (n=5), with dilution linearity demonstrated across 1:2, 1:4, 1:8, and 1:16 dilutions in each matrix. Detection is colorimetric on a pre-coated microplate configured as 12 x 8 well strips, and kit components are stored at +4°C or −20°C as instructed; the kit should not be used past its expiration date.
