This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of rat Receptor Interacting Serine Threonine Kinase 1 (RIP) in tissue homogenates, cell lysates and other biological fluids. The assay uses a 96-well microtiter plate pre-coated with an antibody specific for RIP, and is read on a colorimetric microplate reader at 450 nm.

The kit applies a sandwich immunoassay format: standards and test samples are added to the pre-coated wells, where RIP present in each sample is bound by the immobilized antibody. After incubation and washing, a Biotinylated Anti-RIP Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of the acidic stop solution. The intensity of the yellow colour is proportional to the amount of RIP captured in each well, and the concentration is calculated by referring to the standard curve.

The reported sensitivity is <0.112 ng/ml, with a quantitative range of 0.312–20 ng/ml and a total assay time of 4 hours 30 minutes. The platform is a pre-coated microplate supplied as 12 x 8 well strips. Spike-and-recovery testing (n=5) gave ranges of 80–102% in serum (average 91%), 81–100% in EDTA plasma (average 90%) and 80–89% in heparin plasma (average 84%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 was 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma.
