From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is intended for the in vitro quantitative determination of rat Pyruvate Dehydrogenase Kinase Isozyme 1 (PDK1) in serum, plasma, tissue homogenates, or other biological fluids. The assay is run on a pre-coated 96-well microplate configured as 12 x 8 well strips, with absorbance read at 450 nm on a colorimetric microplate reader.

Standards and samples are added to wells bearing an immobilised PDK1-specific capture antibody, followed by a biotinylated anti-PDK1 detection antibody, HRP-Avidin conjugate, and TMB substrate. The enzymatic reaction yields a blue product that converts to yellow upon addition of acidic stop solution, with the intensity of yellow proportional to the amount of PDK1 captured. Reported quantitative performance includes an assay range of 0.156-10 ng/ml and a sensitivity of 0.057 ng/ml. Total assay time is 4h 30m.
