Rat Pyruvate Dehydrogenase Alpha ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (ELISA) from Antibodies.com designed for the in vitro quantitative determination of rat pyruvate dehydrogenase alpha (PDHA1) in tissue homogenates, cell lysates or other biological fluids. The assay employs a 96-well microplate pre-coated with an antibody specific for PDHA1, a biotinylated anti-PDHA1 detection antibody and an HRP-avidin conjugate, with a TMB substrate read at 450 nm on a colorimetric microplate reader. The density of the yellow colour produced after the acidic stop solution is added is proportional to the amount of PDHA1 captured in each well, and the concentration is calculated from the standard curve.

The kit reports a sensitivity of 0.27 ng/ml and a working standard curve range of 0.781-50 ng/ml, with a total assay time of 4 hours 30 minutes. Spike-and-recovery testing showed serum recovery of 80-102% (average 91%, n=5), EDTA plasma 81-100% (average 90%, n=5) and heparin plasma 80-89% (average 84%, n=5). Linear dilution performance (1:2 to 1:16) is also provided for serum, EDTA plasma and heparin plasma samples.

Components supplied include a pre-coated 96 well microplate (12 x 8 well strips), lyophilized standard (2 vials), detection solutions A and B, a 30X wash buffer, sample dilution buffer, TMB substrate, stop solution and adhesive plate sealers. Components should be stored at +4°C or -20°C as indicated on each item and the kit should not be used past its expiration date.
