Rat Prohibitin ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (ELISA) is designed for the in vitro quantitative determination of rat Prohibitin in serum, plasma or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, in which an antibody specific for Prohibitin is pre-coated onto a microplate. Standards and samples are added to the wells, followed by a Biotinylated Anti-Prohibitin Antibody, an HRP-Avidin conjugate and a TMB substrate. The catalysed reaction yields a blue-coloured product that turns yellow upon addition of an acidic stop solution; the intensity of the yellow colour is proportional to the amount of Prohibitin captured, read at 450 nm in a colorimetric microplate reader and quantified against a standard curve.

Reported performance characteristics include a sensitivity of <0.121 ng/ml and an assay range of 0.312–20 ng/ml. Average recovery values from spiked samples are: serum 91% (range 80–102%, n=5), EDTA plasma 90% (range 81–100%, n=5) and heparin plasma 84% (range 80–89%, n=5). Across serial dilutions (1:2, 1:4, 1:8, 1:16) of five serum, five EDTA plasma and five heparin plasma samples, recovery ranges were respectively 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma. Total assay time is approximately 4 hours 30 minutes, with detection on a colorimetric, pre-coated microplate platform.
