Description

The Antibodies.com sandwich ELISA kit is designed for the in vitro quantitative determination of rat Vitamin K-dependent Protein C in serum, plasma, tissue homogenates, and other biological fluids. The assay runs on a pre-coated microplate and is completed in 4 hours, with a colorimetric readout read at 450 nm on a standard microplate reader.

Performance characteristics include an analytical range of 1.563-100 ng/mL and a sensitivity of <0.938 ng/mL. The antibody pair shows no reported cross-reactivity with other analogs or variants of Vitamin K-dependent Protein C (also known as Anticoagulant protein C, Autoprothrombin IIA, Blood coagulation factor XIV or PROC). Spike-and-recovery validation across matrices (n=10 each) gave ranges and averages of 89%-101% (95%) for serum, 86%-100% (92%) for EDTA plasma and 90%-104% (98%) for heparin plasma.

Serum and plasma samples should be diluted at least 1:2 with Sample Dilution Buffer prior to testing; validated healthy serum and plasma dilutions are 1:100-1:400. The workflow uses a biotinylated detection antibody and HRP-Streptavidin conjugate with TMB substrate, producing a blue product that turns yellow upon addition of the acidic stop solution, with the yellow intensity directly proportional to target concentration. Kit components include lyophilized standard, sample and antibody dilution buffers, biotinylated detection antibody, HRP-Streptavidin conjugate, SABC dilution buffer, TMB substrate, stop solution, wash buffer and plate sealers. The kit is shipped at +4°C; components are stored at +4°C or -20°C as instructed in the protocol, and freeze/thaw cycles should be avoided.
