Rat Presenilin 1 ELISA Kit

Offered by Antibodies.com, this sandwich ELISA is intended for the in vitro quantitative determination of rat Presenilin 1/PS-1 in serum, plasma, tissue homogenates, or other biological fluids. The assay targets Presenilin 1/PS-1 with reactivity confirmed for rat samples.

The kit uses a pre-coated 96-well microplate arranged as 12 x 8 well strips. An antibody specific for Presenilin 1/PS-1 is immobilised on the plate; standards and test samples are added, and the analyte is captured by the immobilised antibody. After washing, a Biotinylated Anti-Presenilin 1/PS-1 Antibody is added, followed by an HRP-Avidin conjugate and TMB substrate. The enzymatic reaction produces a blue product that changes to yellow on addition of acidic stop solution. Absorbance is read at 450 nm on a colourimetric microplate reader and concentration is determined by reference to a standard curve.

The stated assay range is 0.156–10 ng/ml with a sensitivity of 0.061 ng/ml, and total assay time is approximately 4 h 30 min. Spike-recovery averages were 91% for serum (range 80%–102%, n=5), 90% for EDTA plasma (range 81%–100%, n=5), and 84% for heparin plasma (range 80%–89%, n=5). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 was 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma (all n=5). Kit components should be stored at +4 °C or −20 °C as instructed and the kit must not be used past its expiration date.
