A sandwich ELISA from Antibodies.com is designed for the in vitro quantitative determination of rat KCNJ5 (Potassium Inwardly Rectifying Channel Subfamily J, Member 5) in tissue homogenates or other biological fluids. The assay uses a pre-coated 96-well microplate format with colorimetric detection, and absorbance is read at 450 nm using a microplate reader. Total assay time is 4h 30m.

The kit reports a sensitivity of <0.055 ng/ml and an assay range of 0.156–10 ng/ml. Recovery has been evaluated across serum, EDTA plasma, and heparin plasma samples (n=5 per matrix), with average values of 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–100%), and 84% for heparin plasma (range 80–89%). Dilution linearity was tested at 1:2, 1:4, 1:8, and 1:16 with ranges of 87–91%, 87–107%, 74–101%, and 92–97% for serum; 90–105%, 84–101%, 90–101%, and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105%, and 89–91% for heparin plasma.

Kit components should be stored at +4°C or -20°C as instructed, and the product should not be used past its expiration date. The assay follows a sandwich enzyme immunoassay principle in which biotinylated anti-KCNJ5 antibody and HRP-avidin detection produce a colour change upon addition of TMB substrate and acidic stop solution.
