Rat Plasminogen Activator, Urokinase Receptor ELISA Kit

A sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of rat Plasminogen Activator, Urokinase Receptor in serum, plasma or other biological fluids. The assay uses the sandwich enzyme immunoassay technique: an antibody specific for the target is pre-coated onto a 96-well microtiter plate, standards and samples are added and bound by the immobilised antibody, and a Biotinylated Anti-target Antibody is then applied. After addition of an HRP-Avidin conjugate and washing, a TMB substrate is used to visualise the reaction, producing a blue product that turns yellow on addition of acidic stop solution. Absorbance is read at 450 nm and is proportional to target concentration.

Sensitivity is reported as <0.109 ng/ml and the assay range is 0.312-20 ng/ml. Sample recovery data are summarised as follows: serum averaged 91% across a range of 80-102% (n=5); EDTA plasma averaged 90% across a range of 81-100% (n=5); heparin plasma averaged 84% across a range of 80-89% (n=5). Linearity was assessed at dilutions of 1:2, 1:4, 1:8 and 1:16 across the same sample types, with results reported for each matrix.

The colorimetric, quantitative assay is run on a pre-coated microplate arranged as 12 x 8 well strips and the protocol takes approximately 4 hours 30 minutes. Kit components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date. The assay is read on a colorimetric microplate reader.
