Rat Plasminogen Activator Inhibitor 1 ELISA Kit

Manufactured by Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of rat Plasminogen Activator Inhibitor 1 (PAI1) in plasma samples. The assay uses a pre-coated microplate onto which an antibody specific for PAI1 has been immobilized. Standards and test samples are added to the wells, where PAI1 is captured by the immobilized antibody. After washing, a Biotinylated Anti-PAI1 Antibody is added and binds the captured PAI1, followed by an HRP-Avidin conjugate. A TMB substrate solution is then used to visualize the HRP enzymatic reaction; the resulting blue product turns yellow upon addition of acidic stop solution, and absorbance is read at 450 nm in a colorimetric microplate reader. The concentration of PAI1 is calculated by reference to a standard curve.

The assay covers a range of 0.312–20 ng/ml with a sensitivity of <0.128 ng/ml. Total assay time is 4 hours 30 minutes, and detection is colorimetric. Recovery performance in spiked samples (n=5) ranged from 80–102% (average 91%) for serum, 81–100% (average 90%) for EDTA plasma, and 80–89% (average 84%) for heparin plasma. Linearity across sample dilutions of 1:2 to 1:16 spanned 74–108% across the same sample types. Kit components are stored at +4°C or -20°C as instructed, and the product is suitable for use with plasma samples.
