From Antibodies.com, this Rat PTEN ELISA Kit is a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) designed for the in vitro quantitative determination of rat PTEN (Phosphatase and Tensin Homolog) in tissue homogenates, cell lysates, and other biological fluids. The assay uses a 96-well microtiter plate pre-coated with an antibody specific for PTEN. Standards and samples are added to the wells, where PTEN binds to the immobilised capture antibody.

After incubation and washing, a biotinylated anti-PTEN antibody binds the captured PTEN, and an HRP-Avidin conjugate is added to the wells. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of an acidic stop solution. The intensity of the yellow colour is directly proportional to the amount of PTEN captured in each well. The concentration of PTEN is calculated by reading the O.D. absorbance at 450nm in a microplate reader and comparing to the standard curve. The assay has a reported sensitivity of 0.054 ng/ml, a range of 0.156-10 ng/ml, and an assay time of 4h 30m.
