Rat Peroxisome Proliferator Activated Receptor Gamma Coactivator 1 Alpha ELISA Kit

A sandwich ELISA from Antibodies.com designed for the in vitro quantitative determination of rat PGC1 alpha (Peroxisome Proliferator Activated Receptor Gamma Coactivator 1 Alpha) in tissue homogenates, cell lysates or other biological fluids. The assay uses a pre-coated 96-well microplate (12 x 8 well strips) coated with an antibody specific for PGC1 alpha, followed by a Biotinylated Anti-PGC1 alpha Antibody and an HRP-Avidin conjugate. The enzymatic reaction is visualised with TMB substrate, producing a blue product that turns yellow after addition of an acidic stop solution; the density of yellow is proportional to the amount of PGC1 alpha captured in each well and is read at 450 nm in a colorimetric microplate reader.

Reported performance figures include an assay range of 0.156–10 ng/ml and a sensitivity of <0.061 ng/ml, with a total assay time of 4h 30m. Sample recovery has been evaluated across serum (range 83%–100%, average 91%), EDTA plasma (range 90%–98%, average 94%) and heparin plasma (range 89%–102%, average 95%), each with n=5. Kit components comprise the pre-coated microplate (12 x 8 well strips), lyophilised standard (2 vials), Detection Solution A (120 µl) and Detection Solution B (120 µl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and plate sealers (5 adhesive strips). Components are stored at +4°C or –20°C as instructed; the kit should not be used past its expiration date.
