Description

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) supplied by Antibodies.com enables the in vitro quantitative determination of rat P-Selectin Glycoprotein Ligand 1 (PSGL-1) in serum, plasma, tissue homogenates, cell lysates and other biological fluids. The assay employs a pre-coated microplate and the sandwich enzyme immunoassay technique, using a biotinylated anti-PSGL-1 detection antibody and an HRP-avidin conjugate. TMB substrate generates a colour change read at 450 nm on a colorimetric microplate reader, with concentration determined by reference to a standard curve. Sensitivity is 0.227 ng/ml, the assay range is 0.625–40 ng/ml, and the total assay time is 4h 30m.

This kit is suitable for serum, plasma, cell lysates and tissue homogenates. Recovery has been characterised across sample types: serum (n=5) average 91% (range 80–102%), EDTA plasma (n=5) average 90% (range 81–100%), and heparin plasma (n=5) average 84% (range 80–89%). Linearity upon dilution at 1:2, 1:4, 1:8 and 1:16 has been assessed for each matrix, with serum ranging 74–107%, EDTA plasma 79–108% and heparin plasma 82–105% across the dilutions tested. Kit components are stored at +4°C or -20°C as instructed and must not be used past the expiration date.

The product is intended for research use only and is not for diagnostic or therapeutic applications. Standard preparation involves a lyophilised standard reconstituted to a 40 ng/ml stock, then a 1:2 serial dilution series producing standards of 20, 10, 5, 2.5, 1.25 and 0.625 ng/ml, with Sample Dilution Buffer serving as the zero standard. Standards should be used within 15 minutes of preparation. The procedure involves sequential incubation with detection solutions, washing steps, and a final TMB incubation of 15–25 minutes at 37°C in the dark, followed by addition of stop solution.
