Rat Oxoglutarate Dehydrogenase ELISA Kit

This sandwich Enzyme-Linked Immunosorbent Assay from Antibodies.com is designed for the in vitro quantitative determination of rat Oxoglutarate Dehydrogenase (OGDH) in tissue homogenates, cell lysates, or other biological fluids. The assay uses an antibody specific for OGDH that has been pre-coated onto a microplate. Standards and test samples are added to the wells, where OGDH present binds to the immobilized antibody. Following incubation and washing, a Biotinylated Anti-OGDH Antibody is added, followed by an HRP-Avidin conjugate. A TMB substrate solution is then used to visualize the reaction, producing a blue product that changes to yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader, and the concentration of OGDH is calculated by reference to a standard curve.

The reported assay range is 0.156-10 ng/ml with a sensitivity of <0.053 ng/ml. Total assay time is approximately 4 hours 30 minutes. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date. The kit includes a pre-coated microplate, lyophilized standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution, and adhesive plate sealers.

Recovery and dilution linearity have been evaluated across serum, EDTA plasma, and heparin plasma samples. Average recoveries across n=5 samples were 91% for serum (range 80-102%), 90% for EDTA plasma (range 81-100%), and 84% for heparin plasma (range 80-89%). Dilution linearity was assessed at 1:2, 1:4, 1:8 and 1:16 dilutions across the same sample types.
