This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is intended for the in vitro quantitative determination of rat Osteoprotegerin in serum, plasma, tissue homogenates or other biological fluids. An antibody specific for Osteoprotegerin is pre-coated onto the microplate; standards and samples are added, washed, and detected using a Biotinylated Anti-Osteoprotegerin Antibody followed by an HRP-Avidin conjugate and TMB substrate. The resulting colour change is proportional to Osteoprotegerin concentration and is read at 450 nm on a colorimetric microplate reader.

The kit reports a sensitivity of <0.058 ng/ml and a measuring range of 0.156–10 ng/ml, with a total assay time of 4h 30m. Kit components are stored at +4°C or -20°C as instructed and the kit must not be used past its expiration date.

Spike-and-recovery was assessed with n=5 per matrix, giving the following range and average values: serum 80–102% (91%), EDTA plasma 81–100% (90%) and heparin plasma 80–89% (84%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 (n=5) was 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma.
