Rat Octamer Binding Transcription Factor 4 ELISA Kit

This Antibodies.com sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of rat OCT4 in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids.

The assay employs the sandwich enzyme immunoassay technique. An antibody specific for OCT4 is pre-coated onto a 96-well microtiter plate. Standards and test samples are added to the wells, where OCT4 is captured by the immobilised antibody. After washing, a Biotinylated Anti-OCT4 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate solution is then used to visualise the enzymatic reaction, producing a blue-coloured product that changes to yellow after addition of acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader, and OCT4 concentration is calculated against the standard curve. The assay has a stated sensitivity of <0.06 ng/ml and a working range of 0.156–10 ng/ml. Total assay time is approximately 4 hours 30 minutes.

Reported recovery (n=5) is given as serum average 91% (range 80–102%), EDTA plasma average 90% (range 81–100%) and heparin plasma average 84% (range 80–89%). Kit components are to be stored at +4°C or −20°C as instructed, and the kit should not be used past its expiration date.
