Description

This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of rat Nucleobindin 2 (NUCB2) in serum, plasma or other biological fluids. The kit employs a pre-coated microplate platform arranged as 12 x 8 well strips and is reactive with rat samples.

An antibody specific for NUCB2 is pre-coated onto the microplate wells; standards and test samples are added and any NUCB2 present is captured by the immobilised antibody. After washing, a Biotinylated Anti-NUCB2 Antibody binds the captured analyte, followed by addition of an HRP-Avidin conjugate. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that turns yellow after addition of acidic stop solution. The density of yellow is proportional to the amount of NUCB2 captured in each well, and the concentration is calculated by reading the O.D. absorbance at 450 nm in a colorimetric microplate reader and referring to the standard curve. The quoted assay range is 0.312–20 ng/ml, with a sensitivity of <0.102 ng/ml, and total assay time is 4h 30m.

Validation data reported for serum, EDTA plasma and heparin plasma samples (n=5 for each) include average recovery ranges of 80–102%, 81–100% and 80–89% respectively, along with dilution linearity at 1:2, 1:4, 1:8 and 1:16 (serum 87–91% / 87–107% / 74–101% / 92–97%; EDTA plasma 90–105% / 84–101% / 90–101% / 79–108%; heparin plasma 84–95% / 92–105% / 82–105% / 89–91%). Kit components are stored at +4°C or -20°C as instructed and the kit should not be used past its expiration date.
