Rat Nuclear Receptor Coactivator 3 ELISA Kit

A sandwich enzyme-linked immunosorbent assay (ELISA) from Antibodies.com, designed for the in vitro quantitative determination of rat SRC3 in tissue homogenates or other biological fluids. The plate is pre-coated with an antibody specific for SRC3. Standards and test samples are added to the wells, where SRC3 binds to the immobilised antibody. After washing, a Biotinylated Anti-SRC3 Antibody is added and binds the captured SRC3. HRP-Avidin conjugate is then added, and following incubation and washing, TMB substrate solution is used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow on addition of acidic stop solution. Absorbance is read at 450 nm in a microplate reader and the SRC3 concentration is calculated from a standard curve.

The reported assay range is 0.312–20 ng/ml and the sensitivity is <0.115 ng/ml, with a total assay time of 4 hours 30 minutes. Reported validation data show mean recovery in serum of 91% (range 80–102%, n=5), in EDTA plasma of 90% (range 81–100%, n=5), and in heparin plasma of 84% (range 80–89%, n=5). Linearity data are also reported across dilutions from 1:2 to 1:16 for the same sample types. The kit is supplied with a pre-coated 96-well microplate, lyophilised standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution, and adhesive plate sealers, with components stored at +4°C or -20°C as indicated. The kit should not be used past the indicated expiration date.
