Rat Nicotinamide Adenine Dinucleotide Phosphate Oxidase 1 ELISA Kit

From Antibodies.com, this sandwich ELISA kit enables the in vitro quantitative determination of rat NOX1 (Nicotinamide Adenine Dinucleotide Phosphate Oxosphate Oxidase 1) in tissue homogenates, cell lysates or other biological fluids. An antibody specific for NOX1 is pre-coated onto a 96-well microtiter plate. Standards and test samples are added to the wells, where NOX1 is captured by the immobilized antibody. After washing, a Biotinylated Anti-NOX1 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue colour that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of NOX1 captured in each well, and concentration is calculated from the O.D. absorbance at 450 nm using a colourimetric microplate reader.

The quantitative assay range is 0.312–20 ng/ml with a sensitivity of <0.128 ng/ml. Total assay time is approximately 4 hours 30 minutes on a pre-coated microplate platform. Reported average recovery values are 91% in serum (n=5; range 80–102%), 90% in EDTA plasma (n=5; range 81–100%) and 84% in heparin plasma (n=5; range 80–89%). Sample dilution data is reported at 1:2, 1:4, 1:8 and 1:16 across serum, EDTA plasma and heparin plasma. Kit components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
