Rat Neuropeptide S ELISA Kit

A competitive enzyme-linked immunosorbent assay (cELISA) from Antibodies.com, designed for the in vitro quantitative determination of rat Neuropeptide S in serum, plasma or other biological fluids. The assay employs the competitive enzyme immunoassay technique using a 96-well microtiter plate pre-coated with Neuropeptide S antigen. During incubation, Neuropeptide S in samples or standards competes with the immobilised antigen for binding sites on a Biotinylated Anti-Neuropeptide S Antibody; the more analyte present, the less antibody binds to the plate. Following washing, an HRP-Avidin conjugate is added, and TMB substrate is used to visualise the reaction, producing a blue colour that turns yellow upon addition of stop solution. Absorbance is read at 450 nm using a colorimetric microplate reader, with the density of yellow inversely proportional to Neuropeptide S concentration.

The assay has a working range of 15.6-1000 pg/ml and a sensitivity of <4.75 pg/ml, with a total run time of 2 hours 30 minutes. Reported recovery values (n=5) are 80%-102% (average 91%) for serum, 81%-100% (average 90%) for EDTA plasma and 80%-89% (average 84%) for heparin plasma. Dilution linearity across 1:2 to 1:16 dilutions is reported at 87-91%, 87-107%, 74-101% and 92-97% for serum; 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma; and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma. Kit components should be stored at +4°C or -20°C as instructed, and the product should not be used past its expiration date.
