From Antibodies.com, this sandwich ELISA kit is intended for the in vitro quantitative determination of rat Neuronal Pentraxin I (NP-I) in serum, plasma or other biological fluids. The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips, with detection carried out on a colorimetric microplate reader at 450 nm. The standard assay range is 0.156-10 ng/ml with a sensitivity of <0.065 ng/ml, and the total assay time is 4h 30m.

The kit employs a sandwich enzyme immunoassay technique. Standards and samples are added to wells pre-coated with NP-I-specific antibody, washed, then incubated with Biotinylated Anti-NP-I Antibody followed by an HRP-Avidin conjugate. TMB substrate produces a blue colour that turns yellow upon addition of acidic stop solution; absorbance at 450 nm is proportional to NP-I concentration. Recovery has been reported as 80% - 102% (average 91%) in serum (n=5), 81% - 100% (average 90%) in EDTA plasma (n=5), and 80% - 89% (average 84%) in heparin plasma (n=5). Dilutional linearity was evaluated at 1:2, 1:4, 1:8 and 1:16 dilutions (n=5 per sample type). In serum, recoveries at successive dilutions spanned 87-91%, 87-107%, 74-101% and 92-97%; in EDTA plasma, 90-105%, 84-101%, 90-101% and 79-108%; in heparin plasma, 84-95%, 92-105%, 82-105% and 89-91%.

Kit components include a pre-coated 96-well microplate, lyophilized standard, Detection Solution A, Detection Solution B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers. Components should be stored at either +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
