Description

From Antibodies.com, this Sandwich ELISA is designed for the in vitro quantitative determination of rat Neuregulin 1 (NRG1) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay uses the sandwich enzyme immunoassay technique, with an antibody specific for NRG1 pre-coated onto a microplate. Standards and test samples are added to the wells, where NRG1 is captured by the immobilized antibody. After washing, a Biotinylated Anti-NRG1 Antibody binds the captured NRG1, followed by incubation with an HRP-Avidin conjugate. TMB substrate is then added; catalysis produces a blue colour that turns yellow after addition of acidic stop solution. Absorbance is read at 450 nm in a microplate reader and NRG1 concentration is calculated by reference to the standard curve.

The standard curve covers a range of 0.625–40 ng/ml, with a sensitivity of 0.225 ng/ml. Total assay time is 4 hours 30 minutes and detection is colourimetric. Spiking recovery data: serum averaged 91% (range 80%–102%, n=5), EDTA plasma averaged 90% (range 81%–100%, n=5) and heparin plasma averaged 84% (range 80%–89%, n=5). Dilution linearity was assessed at 1:2, 1:4, 1:8 and 1:16 dilutions. For serum (n=5), recoveries were 87%–91% at 1:2, 87%–107% at 1:4, 74%–101% at 1:8 and 92%–97% at 1:16. For EDTA plasma (n=5), recoveries were 90%–105% at 1:2, 84%–101% at 1:4, 90%–101% at 1:8 and 79%–108% at 1:16. For heparin plasma (n=5), recoveries were 84%–95% at 1:2, 92%–105% at 1:4, 82%–105% at 1:8 and 89%–91% at 1:16.

Kit components include a pre-coated microplate, lyophilized standard, detection solution A, detection solution B, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers. Components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
