A sandwich ELISA supplied by Antibodies.com for the in vitro quantitative determination of rat NAT2/AT-2 in tissue homogenates or other biological fluids. The assay uses a pre-coated 96-well microplate (12 x 8 well strips) onto which an antibody specific for NAT2/AT-2 has been immobilised. Standards and test samples are added to the wells, captured NAT2/AT-2 is bound by a biotinylated anti-NAT2/AT-2 antibody, and the signal is developed with an HRP-avidin conjugate and TMB substrate. The colourmetric reaction is read at 450 nm in a microplate reader and the concentration calculated against the standard curve.

Quantitative performance figures quoted by the manufacturer include a sensitivity of <0.218 ng/ml, an assay range of 0.625-40 ng/ml, and a total assay time of 4h 30m. Spike-and-recovery in serum (n=5) is 80-102% (average 91%), in EDTA plasma (n=5) 81-100% (average 90%), and in heparin plasma (n=5) 80-89% (average 84%). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions is 87-107% for serum (n=5), 79-108% for EDTA plasma (n=5) and 82-105% for heparin plasma (n=5).

Kit components are stored at +4°C or -20°C as indicated on the individual labels and should not be used past the expiration date. The protocol is intended for use on a colorimetric microplate reader and is suitable for tissue homogenate samples.
