Rat Myosin Light Chain Kinase ELISA Kit

A sandwich ELISA from Antibodies.com for the quantitative in vitro determination of rat Myosin Light Chain Kinase (MLCK) in tissue homogenates, cell lysates and other biological fluids. The assay is supplied on a pre-coated 96-well microplate arranged as 12 x 8 well strips, with colorimetric readout on a microplate reader.

In the procedure, standards and test samples are added to wells pre-coated with antibody specific for MLCK, capturing the target. After washing, a biotinylated anti-MLCK detection antibody is added, followed by an HRP-avidin conjugate. TMB substrate is then introduced; catalysis produces a blue product that turns yellow upon addition of stop solution. Absorbance is read at 450 nm and the MLCK concentration is interpolated from a standard curve. Total assay time is 4 hours 30 minutes.

Reported performance includes a measuring range of 0.156–10 ng/ml and a sensitivity of 0.057 ng/ml. Spiking recovery in serum averaged 91% (range 80–102%), in EDTA plasma 90% (range 81–100%) and in heparin plasma 84% (range 80–89%), based on n=5 per matrix. The kit comprises a pre-coated microplate, lyophilised standard, biotinylated detection antibody, HRP-avidin conjugate, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, with storage at +4°C or –20°C as indicated per component.
