Rat Myoglobin ELISA Kit

From Antibodies.com, this Rat Myoglobin ELISA Kit is a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) for the in vitro quantitative determination of rat Myoglobin in serum, plasma, tissue homogenates and other biological fluids. The assay uses a pre-coated microtiter plate (12 x 8 well strips) onto which an antibody specific for Myoglobin has been immobilised.

Standards and samples are added to the wells; Myoglobin present binds to the immobilised antibody. After washing, a biotinylated anti-Myoglobin antibody is added, followed by HRP-Avidin conjugate. TMB substrate produces a blue-coloured product that turns yellow on addition of acidic stop solution, and absorbance is read at 450 nm. The assay range is 0.625-40 ng/ml with a sensitivity of 0.264 ng/ml, and the complete protocol takes 4h 30m.

Validation showed mean recovery of 91% from serum (range 80-102%, n=5), 90% from EDTA plasma (81-100%, n=5) and 84% from heparin plasma (80-89%, n=5). Linear dilution testing of the three sample types at 1:2 to 1:16 yielded recoveries between 74% and 108%. Kit components should be stored at +4°C or -20°C as instructed and not used past the expiration date.
