Rat Multidrug Resistance Associated Protein 1 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) intended for the in vitro quantitative determination of rat MRP1 in tissue homogenates, cell lysates, or other biological fluids. Distributed by Antibodies.com, the kit employs an antibody specific for MRP1 pre-coated onto a microplate. Standards and test samples are added to the wells and any MRP1 present is captured by the immobilised antibody. Following washing, a Biotinylated Anti-MRP1 Antibody is added, which binds the captured MRP1, and an HRP-Avidin conjugate is then applied. A TMB substrate solution is used to visualise the HRP enzymatic reaction, producing a blue product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of MRP1 captured per well, and concentration is calculated by reading O.D. absorbance at 450nm in a colorimetric microplate reader against the standard curve.

The assay has a reported sensitivity of <0.118 ng/ml and a working range of 0.312–20 ng/ml, with a total assay time of 4h 30m. Recovery has been characterised in serum, EDTA plasma, and heparin plasma with average recoveries of 91%, 90%, and 84% respectively (n=5 per matrix), and linearity has been demonstrated across sample dilutions of 1:2, 1:4, 1:8, and 1:16. Kit components should be stored at +4°C or -20°C as instructed, and the kit must not be used past its expiration date.
