Rat Mothers Against Decapentaplegic Homolog 1 ELISA Kit

Supplied by Antibodies.com, this ELISA kit is designed for the in vitro quantitative determination of rat Smad1 (Mothers Against Decapentaplegic Homolog 1). It employs a sandwich enzyme immunoassay format intended for use with tissue homogenates, cell lysates, cell culture supernates, or other biological fluids, and is read on a colorimetric microplate reader.

The kit is built around a pre-coated 96-well microplate (12 x 8 well strips) bearing an immobilised antibody specific for Smad1. Standards and samples are added to the captured antibody, followed by a Biotinylated Anti-Smad1 Antibody and an HRP-Avidin conjugate. TMB substrate is then added and the reaction is stopped with an acidic stop solution; absorbance is read at 450 nm, with yellow intensity proportional to Smad1 concentration. The manufacturer states a sensitivity of <0.107 ng/ml within a product range of 0.312–20 ng/ml, and a total assay time of 4 hours 30 minutes.

Spike-and-recovery performance is reported across serum (n=5, range 84–99%, average 91%), EDTA plasma (n=5, range 82–103%, average 92%) and heparin plasma (n=5, range 84–102%, average 93%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 is also characterised for each matrix. Kit components include the pre-coated microplate, lyophilised standard (2 vials), Detection Solution A (120 µl), Detection Solution B (120 µl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and adhesive plate sealers, each with the storage conditions specified by the manufacturer.
