Rat Microsomal Glutathione S Transferase 1 ELISA Kit

A sandwich enzyme-linked immunosorbent assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of rat Microsomal Glutathione S Transferase 1 (MGST1). The assay is compatible with serum, plasma, tissue homogenates, cell lysates, cell culture supernates, and other biological fluids. Standards and samples are added to a pre-coated 96-well microplate arranged as 12 x 8 well strips, where MGST1 is captured by the immobilised antibody. After washing, a biotinylated anti-MGST1 antibody is added, followed by an HRP-avidin conjugate. TMB substrate is then used to visualise the reaction, producing a blue product that turns yellow upon addition of acidic stop solution. The absorbance is read at 450 nm in a colorimetric microplate reader and the MGST1 concentration is calculated against the standard curve.

The quantitative measurement range is 1.56–100 U/L with a sensitivity of <1 U/L. The total assay time is 4 hours 30 minutes. Kit components should be stored at +4°C or -20°C as instructed and not used past the expiration date. Average recovery values from spiked samples (n=5) were 91% (range 80%–102%) for serum, 90% (range 81%–100%) for EDTA plasma, and 84% (range 80%–89%) for heparin plasma, and dilution linearity is reported across 1:2 to 1:16 dilutions for each of the three sample types.

The kit includes a pre-coated 96-well microplate (12 x 8 well strips), lyophilised standard (2 vials), Detection Solution A (120 µl), Detection Solution B (120 µl), Wash Buffer 30X (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml), and five adhesive plate sealers.
