Rat Matrix Metalloproteinase 8 ELISA Kit

Manufactured by Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay is intended for the in vitro quantitative determination of rat Matrix Metalloproteinase 8 (MMP8) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates, and other biological fluids.

The assay uses a 96-well microplate (12 x 8 well strips) pre-coated with an antibody specific for MMP8. Standards and samples are added to the wells, where MMP8 is captured by the immobilised antibody. Following incubation and washing, a biotinylated anti-MMP8 antibody binds the captured MMP8 and an HRP-avidin conjugate is then added. TMB substrate is used to visualise the HRP enzymatic reaction, producing a blue product that changes to yellow upon addition of acidic stop solution. Absorbance is read at 450 nm on a colorimetric microplate reader, and the concentration of MMP8 is calculated by reference to the standard curve. The product range is 78.1-5000 pg/ml and the sensitivity is 33 pg/ml, with a total assay time of 4h 30m.

Reported recovery values for undiluted samples are 80%-102% (average 91%) for serum, 81%-100% (average 90%) for EDTA plasma, and 80%-89% (average 84%) for heparin plasma, each at n=5. Linearity across 1:2 to 1:16 dilutions is documented for serum, EDTA plasma, and heparin plasma (n=5 each). Kit components are stored at +4°C or -20°C as instructed and the kit should not be used past its expiration date.
