Rat Macrophage Derived Chemokine ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of rat Macrophage Derived Chemokine (MDC) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The kit uses a pre-coated 96-well microplate and applies the sandwich enzyme immunoassay technique: an antibody specific for MDC is immobilised on the plate, standards and samples are added, and captured MDC is detected using a Biotinylated Anti-MDC Antibody followed by HRP-Avidin conjugate and a TMB substrate. The enzymatic reaction produces a blue product that changes to yellow upon addition of the acidic stop solution, and the absorbance is read at 450 nm in a microplate reader, with the MDC concentration calculated against the standard curve.

Performance figures given by the manufacturer include an assay range of 23.4-1500 pg/ml, a sensitivity of 8.45 pg/ml, and a total assay time of approximately 4h 30m. Reported average recovery values are 91% for serum (range 80%-102%, n=5), 90% for EDTA plasma (range 81%-100%, n=5) and 84% for heparin plasma (range 80%-89%, n=5). Linearity of dilution has been demonstrated across 1:2, 1:4, 1:8 and 1:16 dilutions for serum, EDTA plasma and heparin plasma. The stated kit components are a pre-coated microplate, a lyophilised standard, Detection Solution A, Detection Solution B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers, to be stored at +4°C or -20°C depending on the component.
