From Antibodies.com, this sandwich ELISA kit is intended for the in vitro quantitative determination of rat Linker for Activation of T-Cell (LAT) in serum, plasma, tissue homogenates, cell lysates, cell culture supernatants or other biological fluids. The assay uses a pre-coated 96-well microplate in a sandwich enzyme immunoassay format: a LAT-specific capture antibody immobilises the analyte on the plate, a biotinylated anti-LAT antibody binds the captured target, and an HRP-avidin conjugate is detected with a TMB substrate, producing a blue colour that turns yellow after addition of the acidic stop solution. Absorbance is read at 450 nm in a microplate reader and the concentration is calculated against the standard curve.

Quantitative performance figures supplied by the manufacturer include a working range of 0.156–10 ng/ml and a sensitivity of 0.036 ng/ml. Average spike-and-recovery (n=5) is 91% for serum (range 80%–102%), 90% for EDTA plasma (range 81%–100%) and 84% for heparin plasma (range 80%–89%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 is reported as 87%–91%, 87%–107%, 74%–101% and 92%–97% for serum; 90%–105%, 84%–101%, 90%–101% and 79%–108% for EDTA plasma; and 84%–95%, 92%–105%, 82%–105% and 89%–91% for heparin plasma (all n=5). The total assay time is stated as 4h 30m, and components are stored at +4°C or −20°C as instructed; kits must not be used past the stated expiration date.
