Rat Left/Right Determination Factor 1 ELISA Kit

This sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of rat Left/Right Determination Factor 1 (Lefty). It is reactive with rat samples and is intended for use with serum, plasma, tissue homogenates or other biological fluids, and falls within the Assays and Kits product category.

The assay employs the sandwich enzyme immunoassay technique. A Lefty-specific antibody is pre-coated onto a 96-well microplate, and standards or samples are added so that Lefty is captured by the immobilised antibody. After washing, a biotinylated anti-Lefty antibody binds the captured analyte, followed by HRP-avidin conjugate addition and further incubation. The reaction is visualised using TMB substrate solution, which produces a blue-coloured product that changes to yellow after addition of acidic stop solution; absorbance is read at 450 nm in a colorimetric microplate reader. The assay is quantitative and colourimetric, run on a pre-coated microplate of 12 x 8-well strips with a total assay time of approximately 4 hours 30 minutes. Kit components should be stored at +4°C or -20°C as instructed.

Manufacturer-quoted performance figures include an assay range of 0.156–10 ng/ml and a sensitivity of 0.056 ng/ml. Reported sample recovery values are: serum (n=5) 80%–102% with an average of 91%; EDTA plasma (n=5) 81%–101% with an average of 91%; and heparin plasma (n=5) 80%–89% with an average of 84%. Dilution linearity is also reported across 1:2, 1:4, 1:8 and 1:16 dilutions for each of these sample types.
