Rat Klotho ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) kit is intended for the in vitro quantitative determination of rat Klotho in serum, plasma, tissue homogenates and other biological fluids. The assay uses a pre-coated 96-well microtiter plate, with standards and samples added to wells where Klotho is captured by the immobilised antibody, followed by a biotinylated anti-Klotho antibody and HRP-avidin conjugate. TMB substrate produces a blue product that turns yellow upon addition of acidic stop solution, with absorbance read at 450 nm on a colorimetric microplate reader.

Reported performance includes a sensitivity of <0.113 ng/ml, a product range of 0.312-20 ng/ml, and a total assay time of 4h 30m. Validation data for serum, EDTA plasma, and heparin plasma (n=5 each) showed ranges of 80% - 102%, 81% - 99%, and 80% - 89% with corresponding averages of 91%, 90%, and 84%. Dilution linearity at 1:2, 1:4, 1:8, and 1:16 dilutions was reported as 87-91%, 87-107%, 74-101%, and 92-97% for serum; 90-105%, 84-101%, 90-101%, and 79-108% for EDTA plasma; and 84-95%, 92-105%, 82-105%, and 89-91% for heparin plasma.

Supplied components include a pre-coated 96-well microplate, lyophilised standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution, and plate sealers, with storage at +4°C or -20°C as instructed in the protocol manual.
