Rat Ki-67 Protein ELISA Kit

From Antibodies.com, this sandwich ELISA enables the in vitro quantitative determination of rat Ki67 in tissue homogenates, cell lysates and other biological fluids. The assay uses an antibody specific for Ki67 pre-coated onto a 96-well microplate, with a biotinylated anti-Ki67 detection antibody and an HRP-avidin conjugate. TMB substrate produces a colour change read at 450 nm in a microplate reader, and Ki67 concentration is calculated against a standard curve.

The assay has a working range of 0.312-20 ng/ml and a sensitivity of <0.117 ng/ml, with a total assay time of 4h 30m on a pre-coated 12 x 8 well strip microplate. Detection is colourimetric and components are stored at +4°C or -20°C as instructed. Manufacturer validation reports recovery (n=5) of 83%-100% (average 91%) in serum, 91%-100% (average 95%) in EDTA plasma and 91%-101% (average 96%) in heparin plasma. Dilution linearity at 1:2, 1:4, 1:8 and 1:16 was 78-110%, 84-96%, 90-101% and 77-107% for serum (n=5); 93-96%, 89-109%, 72-106% and 90-95% for EDTA plasma (n=5); and 84-104%, 82-101%, 95-101% and 84-110% for heparin plasma (n=5).

Components supplied are a pre-coated 96-well microplate (12 x 8 well strips), lyophilized standard (2 vials), Detection Solution A (120 µl), Detection Solution B (120 µl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and 5 adhesive plate sealers.
