Rat Interleukin 7 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (ELISA) from Antibodies.com, this kit is designed for the in vitro quantitative determination of rat IL-7 in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids.

The assay uses a pre-coated microplate with an antibody specific for IL-7. Standards and samples are added to the wells, where IL-7 binds to the immobilized capture antibody. After incubation and washing, a biotinylated anti-IL-7 antibody binds the captured IL-7, followed by HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue colour that changes to yellow after addition of acidic stop solution. The intensity of yellow is proportional to the amount of IL-7 captured in each well. Concentration is calculated by reading the absorbance at 450 nm on a colorimetric microplate reader and referring to the standard curve. Total assay time is 4 hours 30 minutes.

The assay covers a product range of 15.6–1000 pg/ml with a sensitivity of <6.5 pg/ml. Reported average recoveries are 91% for serum (n=5, range 80–102%), 90% for EDTA plasma (n=5, range 81–100%), and 84% for heparin plasma (n=5, range 80–89%). Kit components should be stored at +4°C or -20°C as instructed in the protocol, and the kit should not be used past its expiration date.
