Rat Interleukin 34 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (ELISA) from Antibodies.com, designed for the in vitro quantitative determination of rat Interleukin 34 (IL-34) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The kit uses a pre-coated 96-well microplate arranged as 12 x 8 well strips, with a colorimetric readout measured at 450 nm on a standard microplate reader.

The assay reports a sensitivity of 5.8 pg/ml and a standard curve range of 15.6-1000 pg/ml, with a total assay time of 4 hours 30 minutes. Standards are prepared by reconstituting the lyophilized standard to a 1000 pg/ml stock and producing a 1:2 dilution series down to 15.6 pg/ml. Detection uses a Biotinylated Anti-IL-34 antibody, an HRP-Avidin conjugate and a TMB substrate; the reaction is terminated with an acidic stop solution, producing a yellow colour whose intensity is proportional to the IL-34 concentration in each well.

Sample recovery data are reported for serum (average 91%, range 83%-100%), EDTA plasma (average 94%, range 91%-98%) and heparin plasma (average 94%, range 90%-99%), with linearity demonstrated across 1:2, 1:4, 1:8 and 1:16 dilutions for each matrix. Kit components include the pre-coated microplate, lyophilized standard, Detection Solution A, Detection Solution B, a 30X wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, stored at +4°C or -20°C as instructed.
