Supplied by Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of rat Interleukin 23 Receptor (IL-23R) in tissue homogenates, cell lysates, and other biological fluids [S1]. The assay runs on a pre-coated 96-well microplate arranged as 12 x 8 well strips, with absorbance read on a colorimetric microplate reader at 450 nm [S1].

The standard curve covers 0.156–10 ng/ml and the kit's sensitivity is reported as <0.042 ng/ml [S1]. The full assay takes 4 hours 30 minutes [S1]. In the procedure, an antibody specific for IL-23R is pre-coated onto the plate; standards and samples are added and IL-23R present in each sample is bound by the immobilized antibody [S1]. After washing, wells are incubated with a Biotinylated Anti-IL-23R Antibody followed by HRP-Avidin conjugate [S1]. TMB substrate is then added, producing a blue product that turns yellow upon addition of acidic stop solution; the absorbance at 450 nm is proportional to IL-23R concentration and is quantified against the standard curve [S1].

Reported mean recovery is 91% in serum (range 83–100%, n=5), 94% in EDTA plasma (91–98%), and 94% in heparin plasma (90–99%) [S1]. Dilution linearity across 1:2 to 1:16 is broadly maintained: serum 78–104%, EDTA plasma 81–109%, and heparin plasma 74–105% [S1]. Kit components include pre-coated strips, lyophilized standard, detection solutions A and B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution, and plate sealers, stored between +4°C and -20°C as instructed [S1].
