A sandwich Enzyme-Linked Immunosorbent Assay from Antibodies.com designed for the in vitro quantitative determination of rat IL-17A in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay employs the sandwich enzyme immunoassay technique, using a pre-coated 96-well microplate (12 x 8 well strips) onto which an antibody specific for IL-17A has been immobilised. Standards and test samples are added to the wells and IL-17A present in each sample is bound by the immobilised antibody. Following incubation and washing, a Biotinylated Anti-IL-17A Antibody is added, which binds the captured IL-17A in each well. An HRP-Avidin conjugate is then added and the plate incubated. After further washing, a TMB substrate solution is used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution.

The density of yellow is proportional to the amount of IL-17A captured in each well. The concentration of IL-17A is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve. The kit has a reported sensitivity of <31 pg/ml and a product range of 78.1-5000 pg/ml, is read on a colorimetric microplate reader, and has a total assay time of 4h 30m. Kit components are stored at +4°C or -20°C as instructed, and the product should not be used past its expiration date.

Recovery performance has been evaluated across sample matrices, with reported average values of: Serum (n=5) 91% across a range of 83%-100%; EDTA Plasma (n=5) 96% across a range of 90%-103%; Heparin Plasma (n=5) 95% across a range of 89%-102%. Dilution linearity has also been examined at 1:2, 1:4, 1:8 and 1:16 dilutions for serum, EDTA plasma and heparin plasma samples.
