Rat Interleukin 10 ELISA Kit

From Antibodies.com's Assays and Kits range, this sandwich ELISA is intended for the in vitro quantitative measurement of rat Interleukin 10 (IL-10). The kit is reactive with rat samples and accepts serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids.

The assay uses the sandwich enzyme immunoassay format on a pre-coated 96-well microplate. An antibody specific for IL-10 is immobilised on the plate; standards and samples are added and any IL-10 present is captured. After washing, a Biotinylated Anti-IL-10 Antibody is added, followed by an HRP-Avidin conjugate. A TMB substrate solution is then added to visualise the enzymatic reaction, producing a blue product that turns yellow upon addition of the stop solution. The density of yellow is proportional to the amount of IL-10 captured, and the concentration is calculated from the O.D. absorbance read at 450 nm on a colorimetric microplate reader.

According to the manufacturer, the product range is 78.1–5000 pg/ml and the sensitivity is <27 pg/ml, with a total assay time of 4h 30m. Reported recovery across five samples averages 91% for serum (range 83–100%), 96% for EDTA plasma (range 90–103%) and 95% for heparin plasma (range 89–102%). Linearity has been demonstrated at dilutions of 1:2, 1:4, 1:8 and 1:16 for serum, EDTA plasma and heparin plasma. The kit includes a pre-coated microplate, lyophilised standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, with storage at +4°C or -20°C as instructed.
