Rat Interleukin 1 Beta ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of rat interleukin 1 beta (IL-1 beta) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The 96-well microplate is pre-coated with an antibody specific for IL-1 beta, and analyte is detected using a biotinylated anti-IL-1 beta antibody followed by an HRP-avidin conjugate. Signal is developed with TMB substrate, giving a blue product that turns yellow on addition of acidic stop solution, and read at 450 nm on a colorimetric microplate reader.

The assay has a working range of 15.6–1000 pg/ml with a sensitivity of <5.5 pg/ml, and a total assay time of 4 hours 30 minutes. In spiked samples the kit gave mean recoveries of 91% in serum (range 83–100%, n=5), 96% in EDTA plasma (90–103%, n=5) and 95% in heparin plasma (89–102%, n=5). Dilution linearity was evaluated in serum, EDTA plasma and heparin plasma across 1:2, 1:4, 1:8 and 1:16 dilutions, with values typically remaining within the acceptance windows reported for each matrix.

Reagents supplied include a pre-coated 96 well microplate (12 x 8 well strips), lyophilised standard (2 vials), Detection Solution A (120 µl), Detection Solution B (120 µl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and plate sealers (5 adhesive strips). Components are stored at +4°C or -20°C as indicated, and the kit should not be used past the stated expiration date.
