Rat Interferon Gamma ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of rat Interferon gamma. The kit is intended for serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids, and is read on a colorimetric microplate reader at 450 nm. The pre-coated microplate is supplied as 12 x 8 well strips and the assay can be completed in approximately 4 hours 30 minutes.

The 96-well plate is pre-coated with an antibody specific for Interferon gamma. Standards and samples are added; captured analyte is detected with a biotinylated anti-Interferon gamma antibody followed by HRP-Avidin conjugate and TMB substrate. The enzymatic reaction yields a blue product that turns yellow upon addition of acidic stop solution, with absorbance proportional to the amount of Interferon gamma in each well. The stated sensitivity is <5.3 pg/mL and the working range is 15.6–1000 pg/mL.

Spike-and-recovery values are reported as: serum (n=5) range 80%–102%, average 91%; EDTA plasma (n=5) range 81%–100%, average 90%; heparin plasma (n=5) range 80%–89%, average 84%. Linear dilution of samples at 1:2, 1:4, 1:8 and 1:16 gave recoveries of 87–91%, 87–107%, 74–101% and 92–97% for serum (n=5); 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma (n=5); and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma (n=5). Kit components include a pre-coated 96-well microplate, lyophilized standard, Detection Solution A, Detection Solution B, Wash Buffer (30X), Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers. Store kit components at +4°C or -20°C as instructed; do not use past the expiration date.
