Rat Insulin Like Growth Factor Binding Protein 6 ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of rat IGFBP6 (Insulin Like Growth Factor Binding Protein 6) in serum, plasma or other biological fluids. The workflow uses a pre-coated 96-well microplate (12 x 8 well strips): an antibody specific for IGFBP6 is immobilised on the plate, standards and test samples are added and the analyte is captured, unbound material is washed away, and a Biotinylated Anti-IGFBP6 Antibody followed by an HRP-Avidin conjugate is applied. TMB substrate is then added, producing a blue-coloured product that changes to yellow after addition of the acidic stop solution; the absorbance is read at 450 nm on a colorimetric microplate reader and is directly proportional to the IGFBP6 concentration in each well.

Reported performance figures are a measuring range of 0.156–10 ng/ml and a sensitivity of 0.064 ng/ml, with a total assay time of 4 h 30 m. Spike-and-recovery validation (n=5) returned ranges of 80%–102% (average 91%) in serum, 81%–101% (average 91%) in EDTA plasma and 80%–89% (average 84%) in heparin plasma. Dilution-linearity testing at 1:2, 1:4, 1:8 and 1:16 (n=5) gave 87%–91%, 87%–107%, 74%–101% and 92%–97% in serum; 90%–105%, 84%–101%, 90%–101% and 79%–108% in EDTA plasma; and 84%–95%, 92%–105%, 82%–105% and 89%–91% in heparin plasma.
