Description

This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is intended for the in vitro quantitative determination of rat Huntingtin in serum, plasma, tissue homogenates, or other biological fluids. The assay uses a pre-coated microplate on which an antibody specific for Huntingtin has been immobilised. Standards and test samples are added to the wells and any Huntingtin present is bound by the immobilised antibody. After washing, a Biotinylated Anti-Huntingtin Antibody is added, followed by an HRP-Avidin conjugate. A TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue product that changes to yellow after addition of an acidic stop solution. The density of yellow is proportional to the amount of Huntingtin captured in each well, and the concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to a standard curve.

The kit reports a sensitivity of <0.055 ng/ml and an assay range of 0.156–10 ng/ml, with a total assay time of 4 h 30 min. Detection is performed on a colorimetric microplate reader. Recovery has been characterised for serum (n=5, range 80–102%, average 91%), EDTA plasma (n=5, range 81–100%, average 90%), and heparin plasma (n=5, range 80–89%, average 84%). Dilutional linearity has been evaluated at 1:2, 1:4, 1:8, and 1:16 dilutions across the same sample types. Kit components are intended to be stored at +4°C or –20°C as instructed, and the kit should not be used past its expiration date.
