Rat High Mobility Group Protein 1 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of rat High Mobility Group Protein 1 (HMGB1) in serum, plasma, tissue homogenates and other biological fluids. The assay employs a pre-coated 96-well microplate (12 x 8 well strips) where HMGB1 is captured by an immobilized antibody, detected with a biotinylated anti-HMGB1 antibody and HRP-avidin conjugate, and visualised using TMB substrate. Absorbance is read at 450 nm on a colorimetric microplate reader.

Quantitative performance is defined by a sensitivity of 14.5 pg/ml and an assay range of 31.2-2000 pg/ml, with a total assay time of 4h 30m. Spike-and-recovery testing demonstrated 91% average recovery in serum (n=5, range 80%-102%), 90% in EDTA plasma (n=5, range 81%-100%) and 84% in heparin plasma (n=5, range 80%-89%). Dilution linearity was validated at 1:2, 1:4, 1:8 and 1:16 dilutions across all three sample types, with recoveries generally falling between 74% and 108%.

Kit components include the pre-coated microplate, lyophilized standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, with storage at +4°C or -20°C as instructed.
