Rat Hepcidin ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com enables the in vitro quantitative determination of rat Hepcidin in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The procedure uses a pre-coated 96-well microplate arranged as 12 x 8 well strips, with an antibody specific for Hepcidin immobilised on the plate. Standards and test samples are added to the wells, followed by a Biotinylated Anti-Hepcidin Antibody and an HRP-Avidin conjugate. TMB substrate produces a blue colour that changes to yellow upon addition of acidic stop solution, and absorbance is read at 450nm on a colorimetric microplate reader.

The product measurement range is 0.625-40 ng/ml, with a sensitivity of 0.227 ng/ml. The total assay time is 4h 30m. Reported recovery values, presented as average with range, are: serum 95% (89-101%, n=5), EDTA plasma 96% (91-101%, n=5) and heparin plasma 96% (91-101%, n=5). Linearity upon dilution at 1:2, 1:4, 1:8 and 1:16 has been demonstrated across the same matrices with corresponding recovery ranges.

Kit components include a pre-coated 96-well microplate, lyophilised standard, detection solution A, detection solution B, 30X wash buffer concentrate, sample dilution buffer, TMB substrate, stop solution and plate sealers. Components are stored at +4°C or -20°C as detailed in the kit insert, and the kit should not be used past its expiration date.
