Rat Hepatocyte Nuclear Factor 1 Beta ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, this Rat HNF-1B ELISA Kit is designed for the in vitro quantitative determination of rat HNF-1B in tissue homogenates or other biological fluids, with reactivity confirmed for rat samples. The assay employs the sandwich enzyme immunoassay technique, in which an antibody specific for HNF-1B is pre-coated onto a 96-well microtiter plate. Standards and test samples are added into the wells, and HNF-1B present in each sample is bound by the immobilised antibody. Following incubation and washing, a Biotinylated Anti-HNF-1B Antibody binds the captured HNF-1B, after which an HRP-Avidin conjugate is added. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of HNF-1B captured in each well, and concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.

The assay has a reported sensitivity of 0.058 ng/ml and an assay range of 0.156-10 ng/ml, with a total assay time of 4h 30m. The platform is a pre-coated microplate, and the required instrument is a colorimetric microplate reader. Kit components are stored at +4°C or -20°C as instructed, and the kit should not be used past its stated expiration date. Recovery data for serum, EDTA plasma, and heparin plasma, together with dilution linearity data, are provided with the kit.
