The assay is built around a pre-coated 96-well microplate arranged as 12 x 8 well strips. Standards and samples are added to the wells, where HC-II is captured by the immobilized antibody. After washing, a biotinylated anti-HC-II detection antibody is added, followed by HRP-avidin conjugate and TMB substrate. The yellow colour intensity, read at 450 nm on a colorimetric microplate reader, is directly proportional to the amount of HC-II captured and is referenced against a standard curve.

Designed for plasma samples, the kit offers an assay range of 78.1-5000 ng/ml and a sensitivity of 33 ng/ml, with a total run time of 4 hours 30 minutes. Spike-and-recovery has been assessed in serum, EDTA plasma, and heparin plasma (n=5 per matrix), giving average recoveries of 91%, 90%, and 84% respectively, with linearity data reported across 1:2, 1:4, 1:8, and 1:16 dilutions for each matrix. Kit components are stored at +4°C or -20°C as indicated: the pre-coated plate and lyophilized standard are held at -20°C, while the wash buffer, TMB substrate, stop solution, and plate sealers are stored at +4°C.
